Tyrosine-protein kinase Fgr (EC 2.7.10.2, UniProt P09769, also known as c-fgr, c-src2, c-src-2 proto-oncogene, Gardner-Rasheed feline sarcoma viral (v-fgr) oncogene homolog, p55-Fgr, p58-Fgr, p58c-Fgr, proto-oncogene c-Fgr) is encoded by the FGR (also known as SRC2) gene (Gene ID 2268) in human. Fgr is a member of the Src family of non-receptor protein tyrosine kinases (PTKs). Fgr contains N-terminal myristylation and palmitylation sites, a PTK catalytic domain, as well as SH2 and SH3 domains mediating interactions with phosphotyrosine-containing and proline-rich motifs, respectively. Fgr is a positive regulator of mast cell activation/degranulation and is essential for IgE-mediated passive cutaneous anaphylaxis (PCA) in mice. Fgr appears to act cooperatively with Fyn in the activation of Syk and its downstream targets in a manner that is counteracted by Lyn.
