COUP transcription factor 2 (UniProt: P24468-2, also known as COUP-TF2, COUP-TF II, Apolipoprotein A-I regulatory protein 1, ARP-1, COUP transcription factor II, COUP-TF II, Nuclear receptor subfamily 2 group F member 2) is encoded by the NR2F2 (also known as ARP1, TFCOUP2) gene (Gene ID: 7026) in human. Coup-TF II is a ligand-activated transcription factor that is activated by high concentrations of 9-cis-retinoic acid and all-trans-retinoic acid. Its DNA binding region is localized in amino acids 76-151. COUP-TF II contains two NR C4-type zinc finger domains (aa 79-99 and 115-139). It is widely distributed in human tissues with detectable expression in all tissues examined. Its tissue and cell-specific expression can be modulated both transcriptionally and post-translationally. Through binding to 5 -AGGTCA-3 direct repeats (DRs) with variable spacing, it can modulate the expression of target genes, such as RA receptor beta 2, PEPCK, NGFI-A, and cholesterol 7 -hydroxylase. COUP-TF II action can also be enhanced by interaction with certain coactivators, such as with members of the steroid receptor coactivator family members SRC-1, SRC-2, and SRC-3. Mutations in NR2F2 gene have been linked to multiple types of congenital heart defects characterized transposition of the great arteries, aortic stenosis, and ventricular septal defect.
