Pro-hevein (UniProt: P02877, also known as Major hevein) is encoded by the HEV1 gene in Hevea brasiliensis (Para rubber tree). Pro-hevein is a N-acetyl-D-glucosamine / N-acetyl-D-neuraminic acid binding lectin that can inhibit fungal growth. It is known to cause allergic reaction in human. It contains one barwin domain and one chitin-binding type-1 domain. Inntag 6 IT6 is a small protein domain of Hevea brasiliensis that displays strong structural determinants and does not generate solubility issues in vivo and in vitro. Also, it is not shown to compromise cell fitness when overexpressed and has no detectable functional and localization effects when fused to a variety of target proteins. Unlike the monomeric HA, Flag and MYC tags, Inntag IT6 does not increase the intrinsic variability of critical size in budding yeast cells. Structure of the Inntag IT6 is based on a packed beta-sheet conformation.
