Description
Lethal(2) giant larvae protein homolog 1 (UniProt: Q80Y17, also known as LLGL, Mgl-1, Mlgl) is encoded by the Llgl1 (also known as Llglh) gene (Gene ID: 16897) in murine species. LLGL belongs to the WD repeat L(2)GL family. LLGL is a cortical cytoskeleton protein that is found in a complex involved in maintenance of cell polarity and epithelial integrity. It is involved in the regulation of mitotic spindle orientation, proliferation, differentiation, and tissue organization of neuroepithelial cells. It is generally enriched in developing axons. LLGL stability and function is achieve by its interaction with RanBPM protein, which primarily acts as a scaffolding protein. Its N-terminal region containing SPRY domain is shown to be responsible for binding to RanBPM and this binding functionally extends its half-life by preventing its turnover through ubiquitin-proteasomal pathway. RanBPM is shown enhance tumor suppressor activity of LLGL by inhibiting LLGL-mediated cell proliferation and cell migration. (Ref.: Suresh, B., et al. (2010). J. Biol. Chem. 285(46), 35340-35349).
