CD173 (also known as fucosylated histo-blood group antigens H type 2, Blood group O) and CD174 (also known as Lewis Y or LeY) are developmentally regulated cell surface carbohydrate antigens that are expressed to varying degrees on many human carcinomas. They are also expressed on CD34+ hematopoietic progenitor cells, but are absent on mature lymphocytes. Hence, it is suggested that CDS173 and CD174 play a role in homing process of hematopoietic stem cells. Fucosylated glycans are known to be involved in cell-cell and cell-matrix interactions, differentiation, cell proliferation, and apoptosis. CD173 is a monofucosylated H2 where as CD174 is a difucosylated LeY. They are structurally related and have 2 precursor chains (Galbeta1-4GlcNAc-) in common. CD173 is synthesized through the addition of a fucose on type 2 precursor chains by the alpha 1-2 fucosyltransferases. CD174 results from the epistatic interaction on type 2 precursor chains, of the products of the alpha 1-2 fucosyltransferases and alpha 1-3 fucosyltransferases.
