Cytokine receptor-like factor 2 (UniProt: Q9HC73, also known as Cytokine receptor-like 2, IL-XR, Thymic stromal lymphopoietin protein receptor, TSLP receptor) is encoded by the CRLF2 (also known as CRL2, ILXR, TSLPR) gene (Gene ID: 64109) in human. TSLP receptor is a single-pass type I membrane protein that serves as a receptor for thymic stromal lymphopoietin. It forms a functional complex with TSLP and IL7R to stimulate cell proliferation via activation of STAT3 and STAT5. It is primarily expressed in dendrites and monocytes and its expression has also been reported in heart, skeletal muscle, kidney and liver. Its levels are up-regulated in activated peripheral monocytes and THP-1 cells. TSLP receptor is synthesized with a signal peptide (aa 1-21), which is subsequently cleaved off in the mature form. It contains an extracellular domain (aa 23-231), a transmembrane domain (aa 232-252), and a cytoplasmic domain (aa 253-371). It also has a WSXWS motif (aa 200-204) that is essential for proper protein folding and efficient intracellular transport and cell-surface binding. In addition, its Box 1 motif (aa 261-269) is reported to be important for its association with JAKs. Three isoforms of TSLP receptors have been described that are produced by alternative splicing.
