Chromobox protein homolog 1 (UniProt: P83916, also known as HP1Hsbeta, heterochromatin protein 1 homolog beta, heterochromatin protein p25, M31, Modifier 1 protein, p25 beta) is encoded by the CBX1 (also known as CBX) gene (Gene ID:10951) in human. Chromobox protein homolog 1 is a homodimeric nuclear protein that is a component of heterochromatin. It is expressed in all adult and embryonic tissues. It is essential for formation of and gene repression in heterochromatin. It recognizes and binds histone H3 tails methylated at Lys-9, leading to epigenetic repression. Also interacts with lamin B receptor (LBR) to contribute to the association of the heterochromatin with the inner nuclear membrane. The human HP1 homolog CBX1 preferentially recognize H3K9me3 (Peptide binding specificity Kd = 5 mM). However, upon substitution of Threonine 6 to Alanine reduces its affinity significantly (Kd = 45 mM). (Ref.: Kaustov, L., et al. (2011). J. Biol. Chem. 286(1): 521-529).
