B-cell lymphoma/leukemia 11B (UniProt Q99PV8, also known as B-cell CLL/lymphoma 11B, BCL-11B, COUP-TF-interacting protein 2, mRit1, Radiation-induced tumor suppressor gene 1 protein) is encoded by the Bcl11b (also known as Ctip2, Rit1) gene (Gene ID 58208) in murine species. CTIP2/CBL11B is a C2H2 zinc finger protein that binds GC-rich response elements and is reported to associate with a variety of cofactors, including the corepressor complexes NuRD, Sirt1, histone acetyltransferase (HAT) p300, functioning both as a transcriptional repressor and activator. CTIP2/CBL11B is required at multiple developmental checkpoints for T cell commitment and maintenance. CTIP2/CBL11B expression is initiated at double-negative 2 (DN2) stage of T cell development and is maintained in mature T lymphocytes, with lower levels found in NK cells, but not in B cells, myeloid or dendritic cells. Bcl11b-/- mice die shortly after birth due to a blockade of thymocyte development at the DN2 stage. Bcl11b-deficient DN2 thymocytes are highly proliferative and retain the potential to differentiate into NK cells and myeloid lineages, but not mature T-cells.
