Transmembrane 4 L6 family member 1 (UniProt: P30408, also known as Membrane component chromosome 3 surface marker 1, Tumor-associated antigen L6, TM4SF1) is encoded by the TM4SF1 (also known as M3S1, TAAL6) gene (Gene ID: 4071) in human. TM4SF1 is a multi-pass membrane protein of the L6 tetraspanin family with tetraspanin topology. It has three cytoplasmic domains, four transmembrane domains, and two extracellular domains. It is highly expressed in lung, breast, colon, and ovarian carcinoma. It is highly and selectively expressed on the plasma membranes of tumor cells and on tumor-associated endothelium. It regulates cell motility and proliferation. In PANC-1 and AsPC-1 cells, TM4SF1 is reported to mediate the expression and activities of matrix metallopeptidase 2 (MMP2) and matrix metallopeptidase 9 (MMP9), which are considered as the major regulators of extracellular matrix degradation to increase the cells migration and invasion. Clone 8G4 is reported to be highly effective in reducing blood vessel formation in Matrigel plugs and is shown to destroy PC3 cells and vascular components incorporated into the Matrigel mixture.
