Death-inducer obliterator 1 (UniProt Q9BTC0, also known as DATF-1, Death-associated transcription factor 1, DIO-1, hDido1) is encoded by the DIDO1 (also known as C20orf158, DATF1, KIAA0333) gene (Gene ID 11083) in human. The DIDO1 gene encodes four alternatively spliced variants (DIDO1/isoform 2, DIDO2/isoform 1, DIDO3/isoform 4, isoform a/isoform 3) implicated to be involved in apoptosis and development. The longest and most widely expressed DIDO3 is assigned as the canonical isoform. DIDO3 is a nuclear protein that forms part of the spindle assembly checkpoint (SAC) and is necessary for correct chromosome segregation in somatic and germ cells. Dido3-knockout compromises differentiation of murine embryonic stem cells (mESCs) in vitro and of epiblast cells in vivo, resulting in early embryonic death at around day 8.5 of gestation. On the other hand, exogenous expression of Dido1 isoform in mESCs is reported to inhibit LIF withdrawal-induced differentiation, while Dido1 knockdown facilitate mESCs differentiation.
