Beta-catenin-interacting protein 1 (UniProt Q9NSA3, also known as Beta-catenin-interacting protein ICAT, Inhibitor of beta-catenin and Tcf-4) is encoded by the CTNNBIP1 (also known as ICAT) gene (Gene ID 56998) in human. Inhibitor of beta-catenin and TCF (ICAT) negatively regulates T-cell factor/lymphoid enhancer factor (TCF/LEF)-beta-catenin transcriptional activity by competing against TCF/LEF for beta-catenin binding. ICAT gene knockout leads to premature death of newborn mice with gut, kidney, and craniofacial anomalies, implicating an important role of ICAT in epithelial establishment/maintenance and neural crest cell differentiation. The motility of melanoma cells through 3D matrices in vitro relies on two interconvertible modes of movements. ICAT is reported to suppress the expression of developmentally downregulated 9 (NEDD9) of Wnt/beta-catenin signaling pathway in human Lu1205 melanoma cells, resulting in a shift from the Rac-dependent mesenchymal movement to the Rho/ROCK-dependent amoeboid motility in vitro and enhanced colonization of transplanted Lu1205 in the lungs of nude mice in vivo.
