Tetranectin (UniProt: P05452, also known as TN, C-type lectin domain family 3 member B, Plasminogen kringle 4-binding protein) is encoded by the CLEC3B (also known as TNA) gene (Gene ID: 7123) in human. Tetranectin is a tetrameric plasminogen-binding protein that is synthesized with a signal peptide (aa 1-21), which is subsequently removed in the mature form that is composed of three identical, noncovalently linked subunits each of 181 amino acids. The polypeptide consists of a short a-helical segment (aa 47-72) at the amino terminus that is involved in trimerization, followed by a C-type lectin domain (aa 77-198). It is present in both serum and the extracellular matrix and is expressed most abundantly in the lung. The blood concentrations of tetranectin in healthy adults are about 10-12 mg/mL. However, in septic subjects its levels are significantly reduced and supplementation with exogenous tetranectin confers dose-dependent protection against lethal endotoxemia and sepsis. Its levels are shown to be significantly reduced in patients with cardiac failure. Hence, it is also used as a biomarker for risk of heart failure. (Ref.: Chen, W., et al. (2020). Sci. Transl.
