The epithelium of the small intestine is shown to be organized into large numbers of self-renewing crypt-villus units. Crypts (of Lieberkuhn) are moat-like invaginations of the epithelium around the villi and are lined largely with young epithelial cells, which are involved primarily in secretion. At the base of crypts are stem cells that continually divide and are the source of active self-renewal of the epithelium in the crypt and villi. It has been shown that cells in the enterocyte lineage divide several times as they are pushed out of the crypts to actively migrate onto and along the villi. They differentiate further into the mature absorptive cells that express all the transport proteins and enzymes. Clone E5D10 is highly useful in identifying and isolating Lgr5+ and Bmi1+ intestinal stem cell populations. Although this antibody displays pan-epithelial staining pattern within the small intestine and down along the length of the intestine, it shows more robust staining on the surface of the differentiated epithelial cells on villi and the upper crypt within the small intestine. In the colon this antibody recognizes cells in the colonic cuff epithelium and approximately 2/3 of the way down the colonic crypt. (Ref.: Cleveres, H. (2013).
