Potassium/sodium hyperpolarization-activated cyclic nucleotide-gated channel 4 (UniProt: O70507, also known as Brain cyclic nucleotide-gated channel 3, BCNG-3, HCN4) is encoded by the Hcn4 (also known as Bcng3) gene in murine species. HCN4 is a homo- or hetero-tetrameric multi-pass membrane protein that is detected in a subset of elongated cells in taste buds. It acts as a hyperpolarization-activated ion channel with very slow activation and inactivation. It exhibits weak selectivity for potassium over sodium ions. HCN4 is reported to contribute to the native pacemaker currents in heart that regulates the rhythm of heart beat. It may also contribute to the native pacemaker currents in neurons. HCN4 is activated by cAMP and cAMP binding causes a conformation change that leads to the assembly of an active tetramer and induces channel opening. This channel may mediate responses to sour stimuli.
