Coagulation factor VII (UniProt: P08709, also known as EC: 3.4.21.21, Proconvertin, Serum prothrombin conversion accelerator, SPCA) is encoded by the F7 gene (Gene ID: 2155) in human. Coagulation factor VII is a member of the peptidase S1 family that initiates the extrinsic pathway of blood coagulation. It is serine protease that is synthesized with a signal peptide (aa 1-20) and a propeptide (aa 21-60) and circulates in the blood in a zymogen form. The mature form is a heterodimer of a light and a heavy chain. It is a vitamin K-dependent protease that binds to calcium following enzymatic carboxylation of some glutamate residues. Factor VII is converted to Factor VIIa by the action of Factor Xa, Factor XIIa, Factor IXa, or thrombin by minor proteolysis. In the presence of tissue factor and calcium ions, Factor VIIa converts factor X to factor Xa by cleavage of Arg-|-Ile bond. Factor VIIa also converts factor IX to factor IXa in the presence of tissue factor and calcium. Factor VII deficiency resulting from mutations in F7 gene can lead to hemorrhagic complications.
