Cadherin-6 (UniProt: P55285, also known as Kidney cadherin, K-cadherin) is encoded by the CDH6 gene (Gene ID: 1004) in human. Cadherins are calcium-dependent cell adhesion proteins that preferentially interact with themselves in a homophilic manner in connecting cells. Cadherin-6 is a class II cadherin that is highly expressed in brain and kidney. Its lower expression is also reported in lung, pancreas, and gastric mucosa. It is involved in the morphogenesis of central nervous system and kidney. It is a single-pass type I membrane glycoprotein that is synthesized with a signal peptide (aa 1-18) and a propeptide (aa 19-53) that are subsequently cleaved off to generate the mature form that contains an extracellular domain (aa 54-615), a transmembrane domain (aa 616-636), and a cytoplasmic domain (aa 637-790). It has 5 cadherin domains. Three calcium ions are usually bound at the interface of each cadherin domain and rigidify the connections and thereby imparting a strong curvature to the full-length ectodomain. Cadherin-6 is reported to drive EMT during embryonic development. It acts as a metastasis suppressor molecule and its expression is significantly reduced with an increase in tumor grade and stage in renal cell carcinoma. (Ref.: Gugnoni, M., et al. (2017).
