Profilin-2 (UniProt: P35080, also known as Profilin II) is encoded by the PFN2 gene (Gene ID: 5217) in human. Profilins are actin-binding proteins that serve as key regulators of actin polymerization in cells. Five profilin isoforms have been identified in mammals: Profilins I, IIa, IIb, III and IV. Profilin binds to G-actin in a 1:1 complex and thereby sequesters monomeric actin. Profilin I is highly expressed in all tissues except skeletal muscle, heart, and brain, while profilin II is highly expressed in brain, skeletal muscle, and kidney. Profilin IIa is highly expressed in developing and adult neurons and is involved in correct neurite outgrowth. Profilin IIb appears to be a minor form and it differs from IIa in 13 amino acids in the C-terminal region. Knockdown of both profilins I and II is reported to inhibit neurite outgrowth, possibly impairing actin dynamics by reducing the availability of profilin -G-actin complexes for microfilament growth. In addition to its role in actin polymerization and neurite outgrowth, profilin IIa may also be involved in regulation of synaptic vesicles. It exhibits high affinity for proline-rich peptides and is shown to interact with survival of motor neurons protein (SMN). Mutated forms of SMN (Y272G and G279V) are reported to significantly reduce its interaction with profilin IIa. (Ref.: Sharma, A., et al. (2005). Exp. Cell Res. 305, 195-197, Witke, W., et al. (1998).
