Dachshund homolog 1 (UniProt: Q9UI36, also known as Dach1) is encoded by the DACH1 (also known as DACH) gene (Gene ID: 1602) in human. Dach1 is a transcription factor that is involved in regulation of organogenesis. Four isoforms of Dach1 have been reported that are produced by alternative splicing. It is shown to be widely expressed. Isoform 2 is found in brain, heart, kidney, liver, leukocytes and spleen. Isoform 3 is found in liver and heart and isoform 4 is found in spleen. Clone 1F6.1 exhibits reactivity with all four isoforms. Dach1 is reported to act as regulator of SIX1, SIX6, and probably SIX5. It also acts as a a corepressor of SIX6 in regulating proliferation by directly repressing cyclin-dependent kinase inhibitors, including the p27Kip1 promoter. Dach1 is shown to Inhibit TGF-beta signaling through interaction with SMAD4 and NCOR1. It binds to chromatin DNA via its DACHbox-N domain, which forms a structure containing a DNA binding motif similar to that of the forkhead/winged helix domain. PELP1 (proline, glutamic acid, and leucine rich protein 1) serves as a Dach1-binding protein and the carboxyl terminus of Dach1 is required for binding to PELP1. Dach1 is reported to prevent the proliferation of cancerous cells due to being a natural repressor of estrogen receptors (ER) in the normal function of mammary tissue.
