Killer cell lectin-like receptor subfamily B member 1 (UniProt: Q0ZUP0, also known as Immunoreceptor NKR-P1E, Killer cell lectin-like receptor subfamily B member 1G, Natural killer cell surface protein NKR-P1G, Natural killer lectin-like receptor 1E) is encoded by the Klrb1 (also known as Klrb1d, Klrb1g, Klrb6, Nkrp1e, Nkrp1g) gene (Gene ID: 689817) in rat. NKR-P1G is a single-pass type II membrane protein with a cytoplasmic domain (aa 1-42), a helical domain (aa 43-63), and an extracellular domain (aa 64-214). NKR-P1G is expressed by subsets of lymphocytes with highest prevalence in gut and liver mucosa. The number of NKR-P1G+ cells are shown to increase with age and its expression level is modulated by cytokines, with an increase induced by inflammatory cytokines. It is shown to be mainly expressed by Ly49s3+ cells. It is reported to inhibit cytotoxicity and calcium responses in IL-2-activated NK cells. NKR-P1G is a potent inhibitory receptor for a panel of Clr ligands. The intracellular domain of NKR-P1G contains an immunoreceptor tyrosine-based inhibitory motif that is phosphorylated by Src-kinases upon activation. This allows recruitment of phosphotyrosine phosphatases, which in turn negatively regulates the signaling cascades that is essential for cytotoxicity and cytokine production.
