CCR4-NOT transcription complex subunit 6 (UniProt: Q9ULM6, also known as EC:3.1.13.4, CCR4 carbon catabolite repression 4-like, Carbon catabolite repressor protein 4 homolog, Cytoplasmic deadenylase) is encoded by the Cnot6 (also known as Ccr4, Kiaa1194) gene (Gene ID: 57472) in human species. CNOT6, a catalytic component of the CCR4-NOT complex with in mammals. This complex is ubiquitously expressed and contains at least ten subunits. Subunits CNOT6, CNOT7, and CNOT8 display deadenylase activity and CNOT1, 2, 3, 9, 10, and 11 have regulatory NOT modules. CCR4-NOT complex is reported to limit expression of specific genes through deadenylation of mRNA poly(A) tails, enabling positive selection. CNOT6 is detected both in the nucleus and in cytoplasm. It also serves as a poly(A) nuclease with 3'-5' RNase activity. It has been linked to multiple cellular processes including bulk mRNA degradation, miRNA-mediated repression, translational repression during translational initiation, and general transcription regulation. The deadenylase activity of CCR4-NOT complex has also been implicated in the control of obesity and energy metabolism. In the liver, the deadenylase activity is recruited to the 3 -end of mRNAs of energy metabolism-associated genes and shortens their lifespans and reduces their expression levels. Subunits CNOT 6, 7, and 8 are also shown to participate in the regulation of induced pluripotent stem cell generation and CNOT1, 2, and 3 are involved in maintenance and self-renewal of embryonic stem cells.
