Serine/threonine-protein phosphatase 6 catalytic subunit (UniProt: O00743, also known as EC: 3.1.3.16, PP6C) is encoded by the PPP6C (also known as PPP6) gene (Gene ID: 5537) in human. Protein phosphatase 6 (PP6) is a serine/threonine protein phosphatase that is classified as a type 2A phosphatase family member based on its sequence homology to the catalytic subunit of PP2A. It is a component of a signaling pathway regulating cell cycle progression in response to IL2 receptor stimulation. The PP6 holoenzyme is a heterotrimeric complex of a catalytic subunit, a SAPS domain-containing subunit (PP6R) and an ankyrin repeat-domain containing regulatory subunit (ARS). PP6C is the catalytic subunit of PP6 that has multiple metal binding (manganese) sites and binds 2 manganese ions per subunit. It is ubiquitously expressed in all tissues and highest expression levels are observed in in testis, heart, kidney, brain, stomach, liver and skeletal muscle. PP6C is shown to be upregulated in the glioblastoma and its levels correlate with patient survival time. Silencing of PP6C by small interfering RNA induces sensitivity to IR and delayed release from the G2/M checkpoint.
