Zinc finger and BTB domain-containing protein 4 (UniProt: Q9P1Z0, also known as KAISO-like zinc finger protein 1, KAISO-L1, ZBTB4) is encoded by the ZBTB4 (also known as KIAA1538) gene (Gene ID: 57659) in human. ZBTB4 is an epigenetic regulator that localizes at pericentromeric heterochromatin. It is shown to be an essential component in maintaining genomic stability in mammals. It serves as a transcriptional repressor with bimodal DNA-binding specificity. It represses transcription in a methyl-CpG-dependent manner and binds with a higher affinity to methylated CpG dinucleotides in the consensus sequence 5'-CGCG-3', but can also bind to the non-methylated consensus sequence 5'-CTGCNA-3' (also known as the consensus kaiso binding site). ZBTB4 contains six C2H2-type zinc-finger domains and two proline-rich regions (aa 435-536 and 610-654). Its BTB (Broad-Complex, Tramtrack and Bric a brac) domain is localized in the amino acids 30-152. It is shown to be is frequently downregulated in cancer. ZBTB4 is phosphorylated by HIPK2 (Homeodomain Interacting Protein Kinase 2) and phosphorylation reduces its stability and triggers its degradation in response to DNA damage.
